Thymosin Alpha-1 (Tα1) is a 28-amino acid peptide with the sequence Ac-Ser-Asp-Ala-Ala-Val-Asp-Thr-Ser-Ser-Glu-Ile-Thr-Thr-Lys-Asp-Leu-Lys-Glu-Lys-Lys-Glu-Val-Val-Glu-Glu-Ala-Glu-Asn and a molecular weight of approximately 3,108 Da. Originally isolated from thymic tissue (Thymosin Fraction 5), Tα1 is an N-terminally acetylated peptide that has been extensively studied in immunomodulation, innate immunity, and dendritic cell biology research.

The primary mechanism of action investigated in preclinical models involves Toll-like receptor (TLR) signaling. In dendritic cell (DC) culture models, Tα1 has been shown to activate TLR2, TLR5, and TLR9, leading to MyD88-dependent and TRIF-dependent signaling cascades. This results in NF-κB activation, IRF-7 phosphorylation, and upregulation of type I interferon expression. In vitro maturation assays demonstrate that Tα1-treated monocyte-derived dendritic cells exhibit increased surface expression of MHC class II, CD80, and CD86 co-stimulatory molecules, indicating enhanced antigen-presenting capacity.

T-cell biology represents another critical research area. In mixed lymphocyte reaction (MLR) models, Tα1-matured DCs have demonstrated enhanced capacity to stimulate CD4⁺ T-helper cell proliferation and Th1 polarization, as evidenced by increased IFN-γ and IL-12 production. In isolated T-cell cultures, Tα1 has been shown to promote T-cell differentiation marker expression and modulate the balance between effector and regulatory T-cell populations. NK cell cytotoxicity assays in PBMC preparations have also demonstrated Tα1-mediated enhancement of natural killer cell activity.

Additional research applications include oxidative stress studies, where Tα1 has been shown to upregulate antioxidant enzyme expression including manganese superoxide dismutase (MnSOD) and glutathione peroxidase in macrophage models. Apoptosis research has identified Tα1-mediated modulation of caspase activity and Bcl-2 family protein expression in thymocyte cultures.
