TB-500 is a synthetic peptide corresponding to the active region of Thymosin Beta-4 (Tβ4), a 43-amino acid naturally occurring protein. TB-500 encompasses the actin-binding domain of Tβ4, centered on the sequence LKKTETQ (amino acids 17-23), and has a molecular weight of approximately 4,921 Da. Thymosin Beta-4 and its active fragments have been the subject of extensive preclinical research focused on cell migration, wound repair, and anti-inflammatory mechanisms.

The primary mechanism investigated in preclinical models involves TB-500’s interaction with monomeric G-actin. By sequestering G-actin, the compound modulates the G-actin/F-actin equilibrium, influencing cytoskeletal dynamics critical to cell migration and morphogenesis. In fibroblast and endothelial cell migration assays, TB-500 treatment has been associated with enhanced cell motility and accelerated wound closure in scratch-wound models. This actin-regulatory mechanism distinguishes TB-500 from growth factor-dependent migration pathways.

In vitro angiogenesis studies using endothelial tube formation assays have demonstrated that TB-500 promotes capillary-like network formation. VEGF and angiopoietin expression analysis in treated endothelial cultures suggests TB-500 modulates angiogenic signaling through both direct and indirect mechanisms.
