CJC-1295 without Drug Affinity Complex (no DAC), also designated as Modified GRF(1-29) or Mod GRF 1-29, is a synthetic analog of the first 29 amino acids of human growth hormone-releasing hormone (GHRH). The peptide incorporates four amino acid substitutions at positions 2, 8, 15, and 27 (Ala²→D-Ala², Asn⁸→Gln⁸, Gly¹⁵→Ala¹⁵, Met²⁷→Leu²⁷) to enhance resistance to enzymatic degradation by dipeptidyl peptidase-IV (DPP-IV) and other proteases. In pituitary somatotroph cell cultures, CJC-1295 no DAC activates the GHRH receptor (GHRHR) through Gs-coupled adenylyl cyclase activation, increasing intracellular cAMP levels and promoting GH gene transcription and secretion. Concentration-response studies demonstrate potent GHRHR activation with EC50 values comparable to native GHRH(1-29) but with substantially improved metabolic stability in serum-containing assay conditions.
