CJC-1295 without Drug Affinity Complex (no DAC), also designated as Modified GRF(1-29) or Mod GRF 1-29, is a synthetic analog of the first 29 amino acids of human growth hormone-releasing hormone (GHRH). The peptide incorporates four amino acid substitutions at positions 2, 8, 15, and 27 (Ala²→D-Ala², Asn⁸→Gln⁸, Gly¹⁵→Ala¹⁵, Met²⁷→Leu²⁷) to enhance resistance to enzymatic degradation by dipeptidyl peptidase-IV (DPP-IV) and other proteases. CJC-1295 no DAC has a molecular weight of approximately 3,367.9 Da (peptide portion, excluding the DAC moiety absent in this variant).

The absence of the Drug Affinity Complex is the critical distinction from CJC-1295 with DAC. Without the maleimido-propionic acid linker that enables albumin binding, CJC-1295 no DAC exhibits a shorter half-life in serum-containing cell culture media, more closely modeling pulsatile GHRH signaling dynamics. This pharmacokinetic profile makes CJC-1295 no DAC particularly valuable for research investigating episodic GHRH receptor activation and pulsatile GH release patterns.

In pituitary somatotroph cell cultures, CJC-1295 no DAC activates the GHRH receptor (GHRHR) through Gs-coupled adenylyl cyclase activation, increasing intracellular cAMP levels and promoting GH gene transcription and secretion. Concentration-response studies demonstrate potent GHRHR activation with EC50 values comparable to native GHRH(1-29) but with substantially improved metabolic stability in serum-containing assay conditions.

Research applications include comparison studies with CJC-1295 with DAC to investigate the effects of sustained versus pulsatile GHRHR stimulation on somatotroph signaling kinetics, receptor desensitization, and GH mRNA expression patterns. Combined studies with GHS-R1a agonists (such as Ipamorelin) enable investigation of the synergistic GHRHR/GHS-R1a signaling axis under pulsatile stimulation conditions.

Upon release, Erosyn Labs will supply CJC-1295 no DAC as a lyophilized powder at ≥98% purity by HPLC. Full Certificate of Analysis including mass spectrometry identity confirmation will be provided.
