GLP-1 is a long-acting glucagon-like peptide-1 (GLP-1) receptor agonist with a molecular weight of approximately 4,113.58 Da. Structurally, GLP-1 is based on human GLP-1(7-37) with three key modifications: an Aib (α-aminoisobutyric acid) substitution at position 8 to confer DPP-IV resistance, a Lys34 acylation with a C18 fatty diacid chain via a linker to enable albumin binding, and an Arg34→Lys34 substitution. These modifications collectively extend the pharmacokinetic half-life in albumin-containing systems, making GLP-1 a critical tool for sustained GLP-1 activation research.

In GLP-1 receptor (GLP-1)-expressing cell models, GLP-1 binds with high affinity to the GLP-1, a class B GPCR, activating Gs-mediated adenylyl cyclase/cAMP/PKA signaling. In pancreatic β-cell lines (INS-1, MIN6), GLP-1 treatment has demonstrated glucose-dependent potentiation of insulin secretion, CREB-mediated gene transcription, and enhanced β-cell survival under glucolipotoxic stress conditions. These in vitro findings have established GLP-1 as a standard reference compound in incretin biology research.

Metabolic signaling research extends beyond β-cell models. In hepatocyte cultures, GLP-1 activation by GLP-1 has been associated with reduced lipogenesis gene expression (SREBP-1c, FAS) and enhanced fatty acid oxidation markers. In adipocyte models, GLP-1 has been studied for effects on lipolysis and inflammatory cytokine expression. Neuronal cell models expressing GLP-1 have been used to investigate the compound’s effects on neuroprotective signaling cascades, including Akt/PI3K pathway activation and reduction of endoplasmic reticulum stress markers.

Gastric motility research utilizes GLP-1 in smooth muscle cell preparations and enteric neuron models to investigate GLP-1-mediated modulation of gastric contractility and vagal afferent signaling. Cardiovascular research applications include endothelial cell studies examining GLP-1-mediated effects on nitric oxide production and vascular inflammation markers.
