Semax is a synthetic heptapeptide analog of the adrenocorticotropic hormone (ACTH) fragment 4-10, with the modified sequence Met-Glu-His-Phe-Pro-Gly-Pro. The C-terminal Pro-Gly-Pro extension confers enhanced stability against enzymatic degradation compared to the native ACTH fragment. Semax has a molecular weight of approximately 813.93 Da and has been extensively studied in neuroscience research applications, particularly in models of neuroprotection and cognitive function.
In neuronal cell culture models, Semax has been shown to upregulate brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) expression. Quantitative PCR analysis of treated cortical neuron cultures demonstrates significant increases in BDNF mRNA within hours of compound exposure. This neurotrophic modulation has positioned Semax as a valuable research tool for investigating neuroplasticity mechanisms and neurotrophic signaling cascades.
Preclinical neuroprotection models have utilized Semax extensively. In immune cell models, Semax has demonstrated immunomodulatory properties, including modulation of cytokine profiles in peripheral blood mononuclear cell (PBMC) cultures. Cognitive function research in preclinical models has explored Semax in the context of cholinergic and dopaminergic pathway modulation.
